Regarding HMA in DOS 5.0+, you could use DOS=LOW in config.sys to leave HMA free for win286.

Loading DOS low is fairly pointless as that will cause DOS to use significantly more conventional memory.

I don't recommend to emulate these kind of versions from Windwos 2.x on VirtualBox because you might experience problems. Try other programs, or use a real PC that has such requirements.
